Gene expression profiling was performed on dnCRTC-expressing vs GFP-expressing lung cancer A549 cells, and genes that were differentially regulated by dnCRTC vs GFP were identified. Overall design: There were two groups of A549 cells: one transduced with pMSCV retroviruses expressing GFP control and the other transduced with pMSCV retroviruses expressing dnCRTC [CRTC1(1-55aa)-nls-GFP]. Two biological replicates for each group were included. Cells were independently cultured and RNAs were then harvested for RNA-seq analysis.
本研究针对表达显性负性CRTC(dnCRTC)与表达绿色荧光蛋白(GFP)的肺癌A549细胞进行基因表达谱分析,并鉴定出dnCRTC相较于GFP存在差异调控的基因。整体实验设计:将A549细胞分为两组,一组转导携带GFP对照的pMSCV逆转录病毒,另一组转导携带dnCRTC[CRTC1(1-55aa)-nls-GFP]的pMSCV逆转录病毒。每组均设置两个生物学重复,细胞经独立培养后收集RNA,进行RNA测序(RNA-seq)分析。
