The effects of mechanical ventilation on gene-expression profiles in TGF-b1 induced lung injury

Female mice lungs at the age of 10 to 11 weeks were either subjected to adenoviral vector mediated TGF-b1 (AdTGF-b1) to induce lung injury or an empty vector as healthy control (AdCl). One week later, mice were randomized and either ventiated with a positive end-exspiratory pressure (PEEP) of 2 or 8 cmH2O for up to 4 hours, both at a tidal volume of 10 ml/kg bodyweight and an inspiratory to expiratory time ratio of 1:2. Finally lung tissue was harvested for bulk RNA-sequencing (numbers N6519-N6538). Data were correlated to lung mechanical and structural data. In addition, two groups very included which were not ventilated to investigate the effect of AdTGF-b1 on gene-expression profiles (N5334-N5341).
