Transcriptome of miR-290-295 KO mESCs

In order to identify direct microRNA targets in mouse embryonic stem cells (mESCs), we knocked out microRNA effector genes, profiled their transcriptomes and integrated the data with additional published OMICs data sets. To validate the analysis, we generated miR-290-295 KO mutant cell lines, assessed the transcriptome profile and compared the observed misregulation to the predictions of the analysis. Transcriptome of miR-290-295 KO E14 mESCs were generated in biological duplicates using QuantSeq kit.
为鉴定小鼠胚胎干细胞(mouse embryonic stem cells, mESCs)中的microRNA(微小RNA)直接靶标,我们敲除了microRNA效应基因,对其转录组进行了测序分析,并将所得数据与已发表的其他组学(OMICs)数据集进行整合。 为验证该分析的可靠性,我们构建了miR-290-295基因敲除突变细胞系,对其转录组特征进行了评估,并将观测到的表达失调现象与分析预测结果进行了比对。 本研究采用QuantSeq试剂盒,通过两次生物学重复获取了miR-290-295基因敲除的E14小鼠胚胎干细胞的转录组数据。
