Hypoxia and re-oxygenation are common features in many physiological and pathological processes such as, inflammation and tumorigenesis. Vascular endothelial (VEC) participates in the regulation of a variety of important balances in the body and the regulation of cellular functions, plays an important role in the reaction of organs such as ischemia, hypoxia, and inflammatory immunity. Overall design: HUVEC (Procell Life Science & Technology Co,. Ltid.) were replaced with fresh medium at 37℃ in a humidified atmosphere of 5% CO2 and 95% air which the O2 concentration was about 20% for 12 hours before exposure to hypoxia. Hypoxia cultures (5% O2) were incubated under 5% O2, 5% CO2 and 90% N2 by 3131 Single Tri-gas 184L incubator (Thermo Fisher scientific, Shanghai, China). Hypoxia/re-oxygenation group HUVEC were incubated under hypoxia for 3 hours followed by 1 hour 95% air/5% CO2. HUVEC cultured only in 95% air/5% CO2 were used as the normoxia control group.

缺氧与复氧是诸多生理及病理过程(如炎症发生、肿瘤生成)中的常见特征。血管内皮细胞(Vascular Endothelial Cell, VEC)参与调控体内多种重要生理平衡与细胞功能,在缺血、缺氧及炎症免疫等器官应答过程中发挥关键作用。实验整体设计如下:将人脐静脉内皮细胞(Human Umbilical Vein Endothelial Cell, HUVEC,购自Procell生命科技有限公司)置于含5% CO₂、95%空气(氧气浓度约20%)的湿润培养箱中,于37℃下更换新鲜培养基,预培养12小时后再进行缺氧处理。缺氧组细胞置于5% O₂、5% CO₂及90% N₂的气体环境中,使用3131型Single Tri-gas 184L三气培养箱(赛默飞世尔科技,中国上海)进行培养。缺氧/复氧组人脐静脉内皮细胞先经缺氧处理3小时,随后更换为95%空气/5% CO₂环境培养1小时。仅在95%空气/5% CO₂环境中培养的人脐静脉内皮细胞作为常氧对照组。
