Network analysis reveals strain-dependent response to misfolded tau aggregates

To investigate the effect of wild-derived genetic background on modeling tau aggregation, we injected either AAV-eGFP or AAV-hTauP301L into postnatal day 0 mice to drive widespread expression. We then performed gene expression profiling analysis using data obtained from mRNA-seq from mouse brain cortex of 6 month old hTauP301L- or eGFP-mice. Overall design: Comparative gene expression profiling analysis of mRNA-seq data for eGFP or hTauP301L mice on the following genetic backgrounds: C57BL6/J, CAST/EiJ, PWK/PhJ, and WSB/EiJ (n=4/background/injection/sex=64).

为探究野生来源遗传背景在tau蛋白聚集(tau aggregation)建模中的作用,我们向出生后第0天的小鼠注射腺相关病毒(Adeno-associated virus, AAV)介导的AAV-eGFP或AAV-hTauP301L以实现广泛表达。随后,我们针对6月龄的hTauP301L或eGFP小鼠的大脑皮层mRNA测序(mRNA-seq)数据开展基因表达谱分析。 整体实验设计:对携带C57BL/6J、CAST/EiJ、PWK/PhJ及WSB/EiJ四种遗传背景的eGFP或hTauP301L小鼠的mRNA-seq数据进行对比基因表达谱分析;每类遗传背景、每类注射处理及每个性别的样本量为4,总样本量为64。
