We sought to understand how the RPE confers region-specific disease susceptibility in the retina by examining heterogeneity within human donor RPE. We profiled RPE nuclei from the macular and peripheral retina using joint single-cell RNA and ATAC sequencing. Overall design: RPE nuclei were isolated from macular and peripheral regions of the posterior pole from four healthy human donors. Joint profiling of nuclear chromatin and transcript abundance was performed.
本研究旨在通过探究人类供体视网膜色素上皮(Retinal Pigment Epithelium, RPE)内部的异质性,解析视网膜色素上皮如何赋予视网膜区域特异性的疾病易感性。我们采用单细胞RNA与ATAC联合测序技术,对来自黄斑区与周边视网膜的RPE细胞核进行了谱型分析。
总体实验设计:从4名健康人类供体的视网膜后极部黄斑区与周边区域分离得到RPE细胞核,并完成了细胞核染色质与转录本丰度的联合谱型分析。
