Total RNA was obtained from C4-2 cells with or without SMYD2 silence to perform a high-throughput sequencing to explore the expression profile changes caused by SMYD2 in prostate cancer. AR chromatin immunoprecipitation was conducted using SMYD2-silenced, SMYD2-overexpressed, and parent C4-2 cells. Then, the chromatin immunoprecipitates were extracted to perform high-throughput sequencing to investigate the influence of SMYD2 on AR genomic binding profiles.

本研究从经SMYD2基因沉默或未经SMYD2基因沉默的C4-2细胞中提取总RNA,通过高通量测序(high-throughput sequencing)探究SMYD2对前列腺癌基因表达谱的调控变化。此外,分别以SMYD2基因沉默、SMYD2过表达及亲本C4-2细胞为实验材料,开展针对AR(androgen receptor,雄激素受体)的染色质免疫共沉淀(chromatin immunoprecipitation)实验;随后收集染色质免疫共沉淀复合物并进行高通量测序,以解析SMYD2对AR基因组结合谱的调控作用。
