In Vitro Propagation of XXY Human Klinefelter Spermatogonia Stem Cells; a step towards new fertility opportunities

Human testicular cells of XY and XXY were isolated from cryopreserved tissue in an experimental testicular tissue banking program and propagated in long-term culture for 110 days. Cultured testicular cells were used for cell capture and 10X single cell 3' gene expression sequence library preparation were performed. The resultant library was sequenced using a NovaSeq 6000 SP (100 Cycles).

本研究从一项实验性睾丸组织库项目的冻存组织样本中，分离出XY核型与XXY核型的人类睾丸细胞，并开展了为期110天的长期体外培养扩增。将培养得到的睾丸细胞用于细胞捕获操作，并完成10X单细胞3'端基因表达测序文库的构建。所获文库采用NovaSeq 6000 SP测序仪（100个循环）完成测序。
