We investigated the expression profiles of lncRNAs and mRNAs in peripheral blood mononuclear cells (PBMCs) from subjects with 13 Phlegm-dampness constitution (PDC) and nine balanced Balanced constitution (BC). The differentially expressed lncRNAs (DElncRNAs) and mRNAs (DEmRNAs) were identified by the “limma” method. Goal was to determine whether long non-coding RNAs (lncRNAs) play a regulatory role in metabolic disease in subjects with PDC. Two-condition experiment, BC vs. PDC. Peripheral blood mononuclear cells (PBMCs). Biological replicates:9 subjects with Balanced constitution (BC), 13 subjects with Phlegm-dampness constitution samples (PDC). This experiment was performed technically as a dual channel. Cy3 is used to the label of the PDC and the BC. Besides, RNA from all samples was as reference_RNA. And Cy5 is used to label the reference_RNA in each chip. Normalized log2 ratio (Cy3/Cy5) representing test/reference.

本研究对13例痰湿体质(Phlegm-dampness constitution, PDC)受试者与9例平和体质(Balanced constitution, BC)受试者的外周血单个核细胞(peripheral blood mononuclear cells, PBMCs)中长链非编码RNA(long non-coding RNAs, lncRNAs)和信使RNA(messenger RNA, mRNAs)的表达谱展开分析。采用"limma"方法筛选得到差异表达lncRNAs(DElncRNAs)与差异表达mRNAs(DEmRNAs),旨在明确长链非编码RNA是否在痰湿体质受试者的代谢疾病中发挥调控作用。本实验为双组对照实验,以平和体质组与痰湿体质组为对照,实验样本为外周血单个核细胞。生物学重复设置为:平和体质受试者9例,痰湿体质受试者13例。本实验采用双通道技术开展:Cy3用于标记PDC组与BC组样本,所有样本的RNA均作为reference_RNA,且每张芯片中的reference_RNA均通过Cy5进行标记。最终以归一化后的log2比值(Cy3/Cy5)代表检测样本与参考样本的表达差异。
