We used scRNA-seq to assess the differential effect of Smyd3 ASOs on the transcriptomic profile of different cell types in the TME of MOC1 tumors. Overall design: MOC1 tumors were implanted in the flank of C57BL/6 mice were treated with control (n=3) or Smyd3 ASOs (n=3) for 18 days. Tumors were surgically resected, digested into single-cell suspensions and scRNA-seq was conducted.
本研究采用单细胞RNA测序(single-cell RNA sequencing, scRNA-seq)技术,评估Smyd3反义寡核苷酸(antisense oligonucleotides, ASOs)对MOC1肿瘤肿瘤微环境(tumor microenvironment, TME)内不同细胞类型转录组谱的差异影响。
整体实验设计如下:将MOC1肿瘤接种于C57BL/6小鼠侧腹皮下,分别以对照组(n=3)与Smyd3 ASOs处理组(n=3)干预18天。待干预结束后,手术切除肿瘤组织,将其消化制备为单细胞悬液,随后进行单细胞RNA测序(scRNA-seq)检测。
