Transcriptome Analysis for Antagonistic Molecular Mechanism of Isolated Probiotic Bacteria against Multi Drug Resistant E. coli.. Bacteria

We isolated probiotic bacterial strain TY-11 from yogurts and conducted Transmission Electron Microscope (TEM) experiment, whole genome sequencing, functional and comparative genomic study, biochemical tests, as well as mRNA analysis. From the complete genome sequence and phylogenetic tree constructed with 332 core genes from 262 genomes, the isolate TY-11 was identified as Lactobacillus delbrueckii subsp. indicus. This probiotic killed the multi-drug resistant (MDR) Escherichia coli JW3481-KC within 4 hours, although any kind of distortion was not observed in the surface of the cells of Escherichia coli JW3481-KC under TEM. From this proposed study, we got the insights of potential probiotic features and antimicrobial activities of our isolated probiotic TY-11, which killed Escherichia coli JW3481-KC by secreting acids and hydrogen peroxide in co-culture.
本研究从酸奶中分离得到益生菌菌株TY-11,并开展了透射电子显微镜(Transmission Electron Microscope, TEM)实验、全基因组测序、功能与比较基因组学研究、生化试验以及mRNA分析。基于完整基因组序列以及由262个基因组的332个核心基因构建的系统发育树,分离菌株TY-11被鉴定为德氏乳杆菌印度亚种(Lactobacillus delbrueckii subsp. indicus)。该益生菌可在4小时内杀灭多重耐药(multi-drug resistant, MDR)大肠埃希菌(Escherichia coli)JW3481-KC,但透射电镜观察未发现该大肠埃希菌JW3481-KC的细胞表面存在任何形态畸变。本研究揭示了分离得到的益生菌TY-11所具备的潜在益生菌特性与抗菌活性,该菌株可通过在共培养体系中分泌酸性物质与过氧化氢,杀灭大肠埃希菌JW3481-KC。
