Transcriptome profile of post ovulatory oviduct of pigs

The objective of this study was to investigate the global transcriptomic changes in the porcine oviduct after ovulation and to further investigate the differentially expressed genes (DEGs) associated with the transition from estrus to metestrus phase after ovulation. The RNA-Sequencing of the post ovulatory ampulla (POA) and early luteal ampulla (ELA) tissues were conducted using Illumina NextSeq2000. The R package NOISeq was used to obtain significant differentially expressed genes (DEGs) with probability of differential expression (1-FDR) value ≥ 0.95 and log2 fold change (log2FC) ≥ 1. The DEGs were further explored by bioinformatics analysis for gene ontology, pathway analysis, hub gene analysis and immune system process analysis. The RNA-Sequencing of the post ovulatory ampulla (POA) (n = 3) and early luteal ampulla (ELA) (n = 3) tissues were conducted using Illumina NextSeq2000.

本研究旨在探究排卵后猪输卵管的全局转录组变化,并进一步分析与排卵后发情期向发情后期转变相关的差异表达基因(differentially expressed genes, DEGs)。本研究采用Illumina NextSeq2000平台对排卵后壶腹部(post ovulatory ampulla, POA)与早期黄体期壶腹部(early luteal ampulla, ELA)组织开展RNA测序(RNA-Sequencing)。使用R包NOISeq筛选得到差异表达概率(1-FDR)≥0.95且log2倍变化(log2 fold change, log2FC)≥1的显著差异表达基因。随后通过生物信息学分析手段,对上述差异表达基因进行基因本体(gene ontology, GO)注释、通路分析、核心基因(hub gene)分析以及免疫系统过程分析。本次实验共纳入排卵后壶腹部组织样本3例(n=3)、早期黄体期壶腹部组织样本3例(n=3),均采用Illumina NextSeq2000平台完成RNA测序。
