Spongospora subterranea is a soil borne biotrophic pathogen that causes powdery scab of potato and vectors Potato mop-top virus (PMTV). Obtaining high-quality nucleic acids is difficult due to the durability of the sporosori, intracellular growth, and obligate life style of the pathogen. Genomic DNA was isolated from root galls collected from a field-grown Russet Burbank potato plant in Minnesota State, USA was sequenced by a Minion Nanopore Systems. Sequenced data were De Novo assembled and polished with short reads from previously sequenced isolate by Illumina Miseq. The newly polished assembled genome mapped 100% to the only published draft genome, SSUBK13. The genome was assembled into 346 scaffolds with an estimated size of 31.51 Mb and 10,325 protein-coding genes. The GC content is 45.65% and consistent with other sequenced Plasmodiophorids except Plasmodiophora brassicae. This genome resource will aid in improving our understanding of the genetic architecture of S. subterranea and development of new strategies for managing powdery scab of potato.

马铃薯粉痂菌(Spongospora subterranea)是一种土传活体营养型病原物,可引发马铃薯粉痂病,并作为媒介传播马铃薯帚顶病毒(Potato mop-top virus, PMTV)。由于该病原的休眠孢子囊稳定性较强、可在寄主细胞内生长,且为专性寄生生活方式,获取高质量核酸的难度较大。本研究从美国明尼苏达州田间种植的赤褐色伯班克(Russet Burbank)马铃薯植株的根肿瘿中提取基因组DNA,采用MinION纳米孔测序系统(Minion Nanopore Systems)完成测序。测序所得数据经从头组装后,利用此前通过Illumina MiSeq对该分离株测序获得的短读长数据进行序列抛光校正。经抛光校正后的组装基因组与已发表的唯一一份草图基因组SSUBK13的比对覆盖率达100%。该基因组共组装为346个支架序列,预估总长度为31.51 Mb,包含10325个蛋白编码基因,其GC含量为45.65%,与除芸苔根肿菌(Plasmodiophora brassicae)外的其他已测序根肿菌类群的特征一致。本基因组资源将有助于加深对马铃薯粉痂菌遗传结构的认知,并为研发马铃薯粉痂病防控新策略提供支撑。
