Postweaning multisystemic wasting syndrome in pigs has devastated the swine industry since the 1990s. Porcine circovirus type 2 (PCV2) is the primary cause of this disease. MicroRNAs (miRNAs) are small noncoding RNAs that play important roles in regulating gene expression, especially at the post-transcription level. The expression profiles of miRNAs have been reported in porcine reproductive and respiratory syndrome, porcine parvovirus, and other pig diseases; however, the miRNA expression profiles in pigs infected with PCV2 have not been reported so far. The Laiwu pig (a Chinese indigenous pig breed) has different meat quality, adipogenesis, and disease-resistance from western commercial pig breeds. In this study, four small RNA libraries were constructed from the lung tissue of uninfected and infected Laiwu and Yorkshire/Landrace crossbred (YL) pigs. High-throughput sequencing and bioinformatics were used to determine the abundance and differential expression of miRNAs in the four libraries Overall design: miRNA-seq of four groups as PCV2 infected Laiwu pig (Lw-i), uninfected Laiwu pig (Lw-un), infected western commercial (YL-i) and uninfected western commercial pigs (YL-un)
自20世纪90年代起,猪断奶后多系统衰竭综合征(Postweaning multisystemic wasting syndrome)便对养猪业造成了严重破坏。猪圆环病毒2型(Porcine circovirus type 2, PCV2)是该疾病的主要致病原。微小RNA(microRNAs, miRNAs)是一类小型非编码RNA,在基因表达调控尤其是转录后水平发挥着关键作用。已有研究报道了猪繁殖与呼吸综合征、猪细小病毒及其他猪类疾病中的miRNA表达谱,但目前尚未见猪感染PCV2后的miRNA表达谱相关研究报道。莱芜猪作为中国本土猪品种,在肉质、脂肪生成能力与抗病性方面均与西方商业猪品种存在显著差异。本研究从未感染与感染PCV2的莱芜猪,以及约克夏/长白杂交(YL)猪的肺组织中构建了4个小RNA文库。采用高通量测序与生物信息学分析手段,对4个文库中miRNA的表达丰度与差异表达情况进行了检测。整体实验设计:对4组样本开展miRNA测序,分组分别为PCV2感染莱芜猪(Lw-i)、未感染莱芜猪(Lw-un)、感染西方商业猪(YL-i)以及未感染西方商业猪(YL-un)
