D_1 QC of retroviral vector library with bone marrow genomic DNA

To verify the effective ness of the retroviral cDNA library, bone marrow cells from the C57BL6 mouse were transduced withe library, and the cDNAs were amplified from the genomic DNA. The amplified PCR product was subjected to NGS analysis to evaluate equal contribution from different members of the library.

为验证逆转录病毒cDNA文库的有效性，本研究以C57BL6小鼠的骨髓细胞作为转导靶细胞，将该文库转导至细胞内；随后从基因组DNA中扩增得到cDNA，对扩增获得的PCR产物开展下一代测序（Next-Generation Sequencing, NGS）分析，以此评估文库中不同成员的贡献均衡性。
