Purpose: To compare transcriptomic changes after guvermectin (GV), indole-3-acetic acid (IAA), trans-zeatin (ZT), epibrassinolide (eBL) and gibberellin acid 3 (GA3) treatments in Arabidopsis. Method: 7-day-old MS liquid cultivated Col-0 seedlings were treated by 10 µM GV, 1 µM IAA, 1 µM ZT, 1 µM eBL or 1 µM GA3. Chemicals were added into MS liquid respectively and whole seedling samples were collected after treatment for 5, 30, 180 minutes. In total, we got control group and 5 treatment groups. Three biological replicates were performed. Results: In total, 54 samples were used for RNA sequencing analysis. At least 6 G clean bases were generated for each sample. Comparative analysis identified differences between GV and other plant growth regulators in transcriptional level. Overall design: In this study, transcriptome changes after 10 µM GV, 1 µM IAA, 1 µM ZT, 1 µM eBL or 1 µM GA3 treatment were compared at 5, 30, 180 minutes. Whole seedling samples from control and treated plants were collected for RNA isolation. 18 treatments in triplicate were used in this study.
研究目的:比较经古尔维菌素(guvermectin, GV)、吲哚-3-乙酸(indole-3-acetic acid, IAA)、反式玉米素(trans-zeatin, ZT)、表油菜素内酯(epibrassinolide, eBL)以及赤霉素3(gibberellin acid 3, GA3)处理后拟南芥(Arabidopsis)的转录组变化。实验方法:将生长7天的经MS液体培养基(MS liquid medium)培养的Col-0野生型拟南芥幼苗,分别以10 μM GV、1 μM IAA、1 μM ZT、1 μM eBL或1 μM GA3进行处理。将上述化学试剂分别添加至MS液体培养基中,于处理后5、30、180分钟三个时间点分别收集全幼苗样本。本实验共设置1个对照组与5个处理组,每组设置3次生物学重复(biological replicate)。实验结果:本研究共纳入54份样本进行RNA测序(RNA sequencing)分析,每份样本至少生成6 G的清洁碱基数据。比较分析揭示了GV与其他植物生长调节剂在转录水平上的表达差异。整体实验设计:本研究对比了10 μM GV、1 μM IAA、1 μM ZT、1 μM eBL或1 μM GA3分别处理拟南芥后,在5、30、180分钟三个时间点的转录组变化。收集对照组与处理组植株的全幼苗样本用于RNA提取,本研究共采用18组处理样本(每组设置3次生物学重复)开展实验。
