Comparison of stress and non-stressed ovarian epithelial tumor tissues in nude mice. We injected ovarian cancer cells SKOV3 into nude mice peritoneal cavity. Tumor tissues with good growth were obtained from nude mice, including 4 non-stressed malignant ovarian tumor tissue samples and 3 stressed malignant ovarian tissue samples. High-throughput profiling of miRNA from the collected ovarian tissue samples. We experimentally stressed animals with a physical restraint system that Anil K Sood designed firstly. The physical restraint system is constructed by polymethylmethacrylate which is either transparent or non-transparent, specifically 50ml centrifuge tubes. Sterilization of the experimental equipment was conducted daily. The physical restraint system had approximately 5 holes (u = 4 mm) at the bottom to ensure adequate ventilation. The mice were reared in a room (specific pathogen-free) with controlled temperature, and 12 h/12 h light / dark, respectively.

裸鼠应激与非应激卵巢上皮肿瘤组织的对比分析。本研究将卵巢癌细胞SKOV3注射至裸鼠腹腔内,从裸鼠体内获取生长状态优良的肿瘤组织,其中包含4例非应激性卵巢恶性肿瘤组织样本与3例应激性卵巢恶性肿瘤组织样本。对收集的卵巢组织样本进行miRNA(microRNA)高通量表达谱分析。本实验采用Anil K Sood率先设计的物理束缚系统对实验动物施加应激刺激:该系统由聚甲基丙烯酸甲酯(polymethylmethacrylate,PMMA)制成,可分为透明与非透明两种类型,具体采用50ml离心管制备;实验设备每日均需进行灭菌处理。该物理束缚系统底部开设有约5个孔洞(原文标注孔径为u = 4 mm),以确保充足的通风条件。小鼠饲养于温度可控的无特定病原体(specific pathogen-free,SPF)级动物房内,光照周期设置为12小时明/12小时暗。
