The server version of CLC Assembly Cell was used to assemble de novo Siberian larch (Larix sibirica) genome using the Illumina PE and MP reads. A stepwise approach was developed and used to assemble this large genome (12.03 Gb). It is described in Kuzmin et al. BMC Bioinformatics 2019 20(Suppl 1): 37 ( https://doi.org/10.1186/s12859-018-2570-y). The assembly took 23 days of computing. The number of obtained scaffolds is 11.3 million with a total length of 12.34 Gb. The longest scaffold is 354 Kb, N50 is 6443.

本研究借助CLC Assembly Cell服务器版软件,利用Illumina PE与MP测序读段,对西伯利亚落叶松(Larix sibirica)进行从头(de novo)基因组组装。针对该12.03 Gb的大型基因组,我们开发并应用了分步组装策略,该策略的详细信息已发表于Kuzmin等人的研究:《BMC Bioinformatics》2019年,20(增刊1):37(DOI: 10.1186/s12859-018-2570-y)。本次组装的计算时长总计23天,最终共获得1130万条支架序列(scaffold),总长度为12.34 Gb;最长支架序列长度达354 Kb,组装连续性指标N50为6443。
