We reported the transcriptional effects of gemcitabine, genistein and the combined gemcitaibe/gensitein treatments on two pancreatic cancer cell lines MiaPaCa2 and PANC1. The overall purpose of the study to understand the underlying mechanism of genistein chemoenhancing function. Overall design: Two pancreatic cancer cell lines were treated either with 40nM of gemcitabine (Gem2), 50uM of genistein (Gen) or in combination for 24 hours. RNA was isolated and purified by using Qiagen RNAasy kit. The quality and concentraion of RNA were detected by using bioanalyzer.
本研究报道了吉西他滨(gemcitabine)、染料木黄酮(genistein)以及二者联合处理对两种胰腺癌细胞系MiaPaCa2和PANC1的转录效应。本研究的核心目标为阐明染料木黄酮的化疗增敏潜在作用机制。实验整体设计:将两种胰腺癌细胞系分别以40nM吉西他滨(命名为Gem2组)、50μM染料木黄酮(命名为Gen组)或联合处理24小时。随后采用Qiagen RNAasy试剂盒提取并纯化总RNA,通过生物分析仪(bioanalyzer)检测RNA的质量与浓度。
