A platform for generating expandable, branching and gene-editable ureteric bud organoid from primary mouse and human ureteric bud progenitor cells and human pluripotent stem cells, and its maturation into collecting duct organoid. Overall design: mRNA profiles of mouse ureteric bud (UB) organoids cultured in mUBCM for 5 days, 10 days and 20 days, as well as mouse collecting duct (CD) organoids and adult mouse CD cells purified by FACS from kidneys of 2-month-old Hoxb7-Venus mice.

本平台可从原代小鼠及人输尿管芽(ureteric bud)祖细胞、人多能干细胞中制备可扩增、可分支且基因可编辑的输尿管芽类器官,并可诱导其进一步成熟为集合管类器官。 整体实验设计:对在小鼠输尿管芽条件培养基(mUBCM)中分别培养5天、10天及20天的小鼠输尿管芽(UB)类器官,以及小鼠集合管(CD)类器官和从2月龄Hoxb7-Venus转基因小鼠肾脏中通过荧光激活细胞分选(FACS)纯化得到的成年小鼠集合管(CD)细胞的mRNA表达谱进行检测。
