The goal of this study was to evaluate the transcriptional response of human enteroids/colonoids on transwells to infections (bacterial and rotavirus). Enteroids/colonoids lines C103, C109, D103, D109, I103, I109, J2 and J11 were plated on transwells coated with Matrigel, differentiated, and inoculated (rotavirus (Ito), bacteria or mock) for 6 or 24 hours. Subsequently, total RNA was isolated and paired-end sequencing was performed. Overall design: Enteroid/colonoid mRNA profiles from 48 samples (no technical replicates) were generated by paired-end sequencing.

本研究旨在评估Transwell小室上培养的人肠类器官/结肠类器官(enteroids/colonoids)对细菌及轮状病毒(rotavirus)感染的转录应答。将C103、C109、D103、D109、I103、I109、J2及J11号肠类器官/结肠类器官株接种于包被基质胶(Matrigel)的Transwell小室,经分化诱导后分别接种轮状病毒(Ito株)、细菌或进行模拟感染(mock),感染时长设置为6小时或24小时。随后提取总RNA并开展双端测序(paired-end sequencing)。整体实验设计:通过双端测序(paired-end sequencing)获得了48份样本(无技术重复)的肠类器官/结肠类器官mRNA表达谱。
