Tryptamine is the most important compound of the indoleamine family, and its scaffold has been applied in the design of pharmacological agents with anti-migraine, antibacterial and antitumor effects . tryptamine had a stronger inhibitory effect on high metastatic PCa cell line PC-3 and low metastatic PCa cell line LNCaP compared to the normal prostate epithelial cell line RWPE-1. It also shown to induce cell apoptosis and could markedly inhibit cell migration of PC-3.Metabolic changes induced by tryptamine treatment of PC-3 cells and tumor xenograft mice were also analyzed, with differential metabolites involved in amino acid metabolism and related to prostate cancer development and metastasis identified.
色胺(Tryptamine)是吲哚胺家族中至关重要的化合物,其母核骨架已被应用于具有抗偏头痛、抗菌及抗肿瘤活性的药理活性分子设计。相较于正常前列腺上皮细胞系RWPE-1,色胺对高转移性前列腺癌(PCa)细胞系PC-3与低转移性前列腺癌细胞系LNCaP的抑制作用更强。研究表明,色胺可诱导细胞凋亡,并能显著抑制PC-3细胞的迁移能力。本研究还分析了色胺处理PC-3细胞及肿瘤异种移植小鼠后所引发的代谢变化,鉴定出了参与氨基酸代谢、且与前列腺癌发生及转移相关的差异代谢物。
