The process of orchid protocorm formation during the early stages of symbiotic germination can be classified into five distinct phases: stage 1(A), stage 2(B), stage 3(C), stage 4(D), and stage 5(E). We conducted amplicon sequencing targeting bacterial and fungal regions to investigate the changes in the endophytic microbiome community during different germination stages of Gymnadenia conopsea protocorms under in situ symbiosis. The full-length 16S rRNA sequences amplified by 27F and 1492R primers were selected as bacterial amplicon regions for subsequent sequencing. The fungal amplicons were chosen as the ITS2 regions amplified using ITS86F and ITS4 primers.
兰科植物共生萌发早期阶段的原球茎形成过程可划分为五个明确阶段:阶段1(A)、阶段2(B)、阶段3(C)、阶段4(D)及阶段5(E)。本研究针对细菌与真菌靶区域开展扩增子测序(amplicon sequencing),旨在探究原位共生(in situ symbiosis)条件下,手参(Gymnadenia conopsea)原球茎不同萌发阶段的内生微生物组(endophytic microbiome)群落变化特征。其中,细菌扩增子区域选用27F与1492R引物扩增得到的全长16S核糖体RNA(16S rRNA)序列,用于后续测序;真菌扩增子则选用ITS86F与ITS4引物扩增得到的ITS2区域。
