Bcl11b plays an important role in postnatal dentate gyrus development and adult neurogenesis. To determine its role in adult neurogenesis independant from postnatal development the Bcl11b mutation was induced at the age of 2 months. Microarray analysis was performed on RNA isolated 4 weeks after the induction of the Bcl11b mutation to determine Bcl11b target genes. Control and mutant dentate gyrus (DG) granule cells were isolated by laser capture microdisection and RNA isolated. Transcriptome analysis was performed by BRB-ArrayTools.

Bcl11b在出生后齿状回发育与成年神经发生过程中发挥关键调控作用。为明确其在独立于出生后发育的成年神经发生中的生物学功能,研究人员于2月龄时诱导Bcl11b突变。在Bcl11b突变诱导4周后,提取RNA并开展微阵列分析以筛选Bcl11b的靶基因。通过激光捕获显微切割技术分离对照组与突变组的齿状回(dentate gyrus, DG)颗粒细胞并提取其RNA,随后借助BRB-ArrayTools完成转录组分析。
