Introduction: The microbiome—the community of trillions of microorganisms living in and on the human body—plays a significant role in health and disease, including in pancreatic cancer. Research has increasingly shown that the gut and pancreatic microbiomes may influence the development, progression, and therapeutic intervention of pancreatic cancer. Microbiome dysbiosis (an imbalance in microbial communities) has been linked to an altered immune response in pancreatic cancer. Certain bacteria, for instance, promote the recruitment of immunosuppressive cells in the tumor microenvironment, which can enable cancer cells to evade the immune system.Objectives: Determination of unique phylogenetic operational taxonomic units (OTUs) of microbiome responsible for pancreatic cancer development in mice.Materials & Methods: Isolation of Genomic DNA from fecal matter isolated from healthy mice and mice with subcutaneous PANC-1 human pancreatic cancer xenografts, 16S rDNA PCR and creating amplicon libraries and NG Sequencing, Software analysis of the data generated and identified OTUs of the microbiota.Discussion/Results: A comprehensive data analysis of 16S rDNA sequencing demonstrates increased abundance of several phylogenetic species of bacteria and less abundance of other OTUs of bacteria as per 16S rDNA sequencing data analysis in feces microbiome of mice with pancreatic cancer.
引言:微生物组(microbiome)是定植于人体体表及体内的万亿级微生物群落,其在包括胰腺癌在内的多种健康与疾病状态中发挥重要作用。越来越多的研究表明,肠道微生物组与胰腺微生物组均可影响胰腺癌的发生、进展及治疗干预。微生物组失调(microbiome dysbiosis,即微生物群落失衡)与胰腺癌的免疫应答异常存在关联。例如,某些细菌可促进肿瘤微环境中免疫抑制细胞的募集,从而帮助癌细胞逃避免疫系统的清除。
研究目标:确定小鼠体内与胰腺癌发生相关的微生物组特有系统发育操作分类单元(operational taxonomic units, OTU)。
材料与方法:从健康小鼠以及接种皮下移植人胰腺癌PANC-1细胞的小鼠的粪便样本中分离基因组DNA,通过16S核糖体DNA(16S rDNA)聚合酶链式反应构建扩增子文库并进行下一代测序(NG Sequencing),对生成的数据进行软件分析并鉴定微生物群的操作分类单元(OTU)。
讨论/结果:对16S rDNA测序数据的全面分析表明,胰腺癌模型小鼠粪便微生物组中,多种细菌系统发育物种的丰度显著升高,而其余细菌OTU的丰度则有所降低。
