小鼠胶质母细胞瘤肿瘤祖细胞(Tumor Progenitor Cells, TPCs)中,表皮生长因子受体变体III(Epidermal Growth Factor Receptor Variant III, EGFRvIII)活性高低不同的两组——磷酸化EGFR高表达组(pEGFR-Hi)与磷酸化EGFR低表达组(pEGFR-Lo),在增殖、分化及侵袭能力上存在显著差异。将表达ZsGreen荧光蛋白的TPCs颅内接种至免疫健全FVB小鼠体内,于中位生存期±1天时通过流式细胞术(Fluorescence Activated Cell Sorting, FACS)分离肿瘤细胞,随后进行RNA提取;并对从体内肿瘤中经流式细胞术分离的绿色荧光蛋白阳性肿瘤细胞开展表达分析。
