To investigate transcriptional changes in the heart induced by diet, we performed single-cell RNA sequencing on pooled mouse hearts from normal fat diet (NFD, 10% kcal fat) and high fat diet (HFD, 60% kcal fat) groups using the Parse Biosciences Evercode WT Mini v3 kit. Overall design: Whole hearts from C57BL/6J mice fed either NFD or HFD for 4 weeks (n=3 pooled per group) were processed using the Parse Biosciences Evercode Whole Transcriptome Mini v3 kit for single-cell RNA-seq. Libraries were sequenced on Illumina NovaSeq 6000. Two sublibraries were generated.

为探究饮食诱导的心脏转录组变化,我们使用Parse Biosciences公司的Evercode WT Mini v3试剂盒,对正常脂肪饮食(normal fat diet,NFD,10%热量来自脂肪)组与高脂饮食(high fat diet,HFD,60%热量来自脂肪)组的混合小鼠心脏开展了单细胞RNA测序(single-cell RNA sequencing)。 总体实验设计:对喂食正常脂肪饮食或高脂饮食4周的C57BL/6J小鼠的完整心脏,每组取3只小鼠的心脏混合为一个样本,采用Parse Biosciences Evercode 全转录组Mini v3试剂盒进行单细胞RNA测序建库。建库产物在Illumina NovaSeq 6000测序平台上完成测序,共构建了两个子文库。
