We reported chromatin compaction states within several nucleosomes in HepG2 cells. To assess the local compaction, fromaldehyde-treated chromatin was appled to fractionation via a sedimentation velocity cetrifugation. While less-crosslinked nucleosomes remained in upper fractions as open chromatin, well-crosslinked nucleosomes were sedimented to lower fractions as compact chromatin. Overall design: NGS analyses of DNA flagments prepared from the uppermost and the lowermost fractions of the chromatin fractionation.

本研究报道了HepG2细胞内多个核小体(nucleosomes)的染色质压缩状态。为评估局部染色质的压缩程度,我们对甲醛固定的染色质采用沉降速度离心法(sedimentation velocity centrifugation)进行分级分离。其中,低交联度的核小体以开放染色质形式留存于上层分级组分,而高交联度的核小体则以致密染色质形式沉降至下层分级组分。实验整体设计:对来自染色质分级分离最上层与最下层组分的DNA片段开展下一代测序(Next-Generation Sequencing,简称NGS)分析。
