A multi-omics approach was used to understand how temperature affects the molecular mechanisms of pathogenicity of Lasiodiplodia theobromae at different temperatures. The transcriptome (Illumina TruSeq) of LA-SOL3 strain grown at 25 °C and 37 °C was analysed. For RNA extraction, LA-SOL3 was inoculated into a 250 mL flask containing 50 mL of PDB and incubated at 25 °C or at 37 °C for 4 days. The transcriptome of LA-SOL3 was sequenced by Illumina TruSeq to characterise the differential expression at both temperatures.

本研究采用多组学(multi-omics)策略,旨在探究不同温度条件下可可毛色二孢菌(Lasiodiplodia theobromae)的致病性分子调控机制。针对在25 ℃与37 ℃下培养的LA-SOL3菌株,采用Illumina TruSeq技术开展转录组(transcriptome)分析。实验中,将LA-SOL3菌株接种至装有50 mL马铃薯葡萄糖肉汤(Potato Dextrose Broth,PDB)的250 mL三角瓶中,分别置于25 ℃或37 ℃条件下恒温培养4天以提取RNA。随后通过Illumina TruSeq技术对LA-SOL3的转录组进行测序,以表征两种温度下的基因差异表达特征。
