Determination of the mechanism by which fibrinogen, a central blood coagulation protein, regulates OPC functions and remyelination in the CNS. We used microarrays to determine the genes involved in OPC responses to fibrinogen, which is deposited in the CNS after blood-brain barrier disruption. Primary rat OPCs were stimulated with fibrinogen for 12 hr and subjected to RNA extraction and hybridization on Affymatrix microarrays. Three unstimulated and three fibrinogen-stimulated samples were generated.

本研究旨在解析纤维蛋白原(fibrinogen)——一种核心凝血蛋白——调控中枢神经系统(Central Nervous System, CNS)内少突胶质细胞前体细胞(oligodendrocyte progenitor cells, OPC)功能与髓鞘再生的分子机制。我们采用基因芯片技术,鉴定血脑屏障(blood-brain barrier)破坏后在中枢神经系统内沉积的纤维蛋白原所诱导的少突胶质细胞前体细胞应答相关基因。将原代大鼠少突胶质细胞前体细胞经纤维蛋白原刺激12小时后,提取总RNA并进行Affymatrix基因芯片杂交。本实验共制备3组未经纤维蛋白原刺激的对照样本与3组经纤维蛋白原刺激的实验组样本。
