A wild type strain of Listeria monocytogenes (L. mono) was used for infections. Mice were infected subcutaneously on the footpad with a sublethal dose of 1 × 106 colony-forming units (CFU). Mice were left for 2 days before being sacrificed. Control cells were from either PBS injected mouse or from the uninjected leg of infected animal. Stromal vascular fraction cells from adipose tissue were isolated and single live cells were purified and sequenced using the 10X Genomics Chromium controller. One sample per condition was used.

本实验采用单核细胞增生李斯特菌(Listeria monocytogenes,L. mono)野生型菌株开展感染实验。以1×10^6菌落形成单位(colony-forming units,CFU)的亚致死剂量,对小鼠进行足垫皮下感染。感染两天后,对小鼠实施安乐死。对照细胞分别取自注射磷酸盐缓冲液(Phosphate Buffered Saline,PBS)的小鼠,以及感染小鼠未注射的肢体。从脂肪组织中分离基质血管组分细胞,纯化单个活细胞后,使用10X Genomics Chromium控制器完成测序。每个实验条件仅使用一个样本。
