We conducted chromatin immunoprecipitation for H3K27ac and BRD4, followed by sequencing (ChIP-seq) to investigate super-enhancer (SE) for drug-resistant using cetuximab-resistant cells and control cells. SE analysis revealed that several unique SE-associated genes in cetuximab-resistant cells were identified. Overall design: To investigate the reaction at the genome level in cetuximab-resistant cells, control cells and cetuximab-resistant cells were sent to Active Motif for ChIP-Seq.

我们针对组蛋白H3第27位赖氨酸乙酰化(H3K27ac)与溴域蛋白4(BRD4)开展染色质免疫共沉淀(chromatin immunoprecipitation, ChIP)实验,随后进行测序,即染色质免疫共沉淀测序(ChIP-seq),以研究西妥昔单抗耐药细胞与对照细胞中与耐药性相关的超级增强子(super-enhancer, SE)。超级增强子分析显示,西妥昔单抗耐药细胞中存在多个特异性的超级增强子关联基因。整体实验设计:为在基因组水平探究西妥昔单抗耐药细胞的分子应答特征,我们将对照细胞与西妥昔单抗耐药细胞送至Active Motif公司完成ChIP-seq测序。
