本数据集通过甲基-CpG结合结构域(methyl-CpG-binding domain, MBD)富集结合大规模平行测序(massively parallel sequencing)技术,对45例原发性神经母细胞瘤(neuroblastoma)肿瘤的DNA甲基化组进行了表征分析。
实验整体设计如下:首先将45例原发性肿瘤的DNA进行片段化处理,得到长度约±200 bp的DNA片段;随后使用Diagenode公司的MethylCap试剂盒开展基于MBD的富集实验,继而完成文库制备与多重标记(multiplexing)。输入DNA与捕获得到的DNA均在Illumina Hiseq2000测序平台上进行双端(paired-end)测序。
