To understand regulation mechanism of miR164/targets, transcriptomes of WT and miR164 ovexpression plants with/without heat stress were dissected. 5-day-old seedlings were treated with heat stress at 35oC for 5 days. Total RNAs were isolated for library construction and analyzed by RNA-seq via Solexa platform. Raw sequences were obtained from the Illumina Pipeline softwareIllumina bcl2fastq v2.1.7 and expected to generate 6 G per sample. 为解析miR164与其靶基因的调控机制,本研究对野生型(Wild Type,WT)、miR164过表达植株在有/无热胁迫条件下的转录组进行了分析。选取5日龄幼苗,置于35℃环境中开展为期5天的热胁迫处理。提取总RNA用于构建测序文库,并通过Solexa平台进行RNA测序(RNA-seq)。原始测序序列由Illumina Pipeline软件bcl2fastq v2.1.7获取,预计每个样本可产生6G测序数据。实验整体设计:检测野生型与miR164过表达幼苗在经热胁迫处理与未处理条件下的转录组表达情况。
