Diabetes-resistant and diabetes-prone female New Zealand Obese mice were classified based on liver fat content and early blood glucose concentrations at 10 weeks of age before the onset of T2D. By using transcriptome and DNA methylome analysis of liver samples, we identified early epigenetic alteration in mice and humans which could serve as putative epigenetic biomarkers Overall design: RNA-seq was performed with 5 diabetes resistant (DR) and 6 diabetes prone (DP) animals, DR animals are healthy animals and are considered as control group
在2型糖尿病(Type 2 Diabetes, T2D)发病前,研究人员依据10周龄时的肝脏脂肪含量与早期血糖浓度,对糖尿病抵抗型与糖尿病易感型雌性新西兰肥胖(New Zealand Obese)小鼠完成分组。本研究通过对肝脏样本开展转录组(transcriptome)与DNA甲基化组(DNA methylome)分析,鉴定出了可作为推定表观遗传生物标志物的小鼠及人类早期表观遗传改变。整体实验设计:对5只糖尿病抵抗(diabetes resistant, DR)小鼠与6只糖尿病易感(diabetes prone, DP)小鼠进行RNA测序(RNA-seq),其中糖尿病抵抗小鼠为健康个体,作为对照组。
