We applied RNA-Seq on lung fibroblasts derived from Atm defecient as well as wildtype mice to further understand and reveal a novel pathawy involved in the premture senescence caused by the loss of Atm. 15 samples were analyezd. 3 strains of lung fibroblasts were established from Atm-/- and Atm+/+ C57BL/6 mice (3 for each genotype) and samples were taken for RNA seq at passage 2 and passage 8. As a control , 3 Atm+/+ strains were irradiated with 10 Gy IR to induce stress induced premature senescence (SIPS).

为深入解析并揭示Atm缺失介导的过早衰老相关全新通路,本研究对源自Atm缺陷型及野生型(wildtype)小鼠的肺成纤维细胞开展RNA测序(RNA-Seq)。本次实验共分析15份样本:我们从Atm-/-与Atm+/+ C57BL/6小鼠中分别构建3株肺成纤维细胞系(每种基因型各3株),并分别在第2代和第8代传代时采集样本进行RNA测序。作为对照,我们对3株Atm+/+细胞系施加10 Gy电离辐射(IR)以诱导应激诱导型过早衰老(SIPS)。
