FAM198B is identified as a novel tumor supressor gene in lung adenocarcinoma. This study aim to investigate the effects of the glycoprotein protein, FAM198B, on the biological behavior and global gene expression profiles of lung cancer cells. Expression microarray and bioinformatics analyses indicated that FAM198B mainly regulated ERK-mediated MMP1 pathway in CL1-5 cells, which results in tumor metastasus inhibition. We used microarrays to identify the global gene alterarion in lung cancer CL1-5 cells after ectopic expressing FAM198B Overall design: Etopic expressing FAM198B-V5 and mock control vector in lung cancer CL1-5 cells

FAM198B被鉴定为肺腺癌中的新型肿瘤抑制基因(tumor suppressor gene)。本研究旨在探究该糖蛋白FAM198B对肺癌细胞生物学行为及全基因表达谱的影响。表达微阵列(expression microarray)与生物信息学分析结果表明,FAM198B在CL1-5细胞中主要调控ERK介导的基质金属蛋白酶1(matrix metalloproteinase 1, MMP1)通路,从而抑制肿瘤转移。我们通过微阵列技术鉴定了异位表达FAM198B后的肺癌CL1-5细胞中的全基因表达改变。实验设计:在肺癌CL1-5细胞中异位表达FAM198B-V5及空载对照载体。
