RNA-Seq of explanted murine pancreatic acinar cells from wt, KrasG12D, Pi3kCAH1047R and Mek1dd mice

To investigate the underlying changes during acinar-to-ductal metaplasia induced by different factos, pancreatic acinar cells were isolated from 5 week old wt, KrasG12D, Pi3kCAH1047R and Mek1dd mice. Acinar cells from wt mice were treated with medium (control), Tgfa or IL17a. RNA was isolated immediately after isolation (t=0 days) or after 2-3 days embedded in collagen.

为探究不同因素诱导的腺泡-导管化生（acinar-to-ductal metaplasia）过程中的潜在变化，研究人员从5周龄的野生型（wild type）、KrasG12D、Pi3kCAH1047R及Mek1dd小鼠中分离胰腺腺泡细胞。将野生型小鼠来源的腺泡细胞分别以普通培养基（对照组）、Tgfa或IL17a进行处理。分别在细胞分离后即刻（t=0天），以及于胶原蛋白基质中培养2-3天后提取RNA。
