This study provides P. knowlesi line A1-H.1 bulk mRNAseq data at five time points in the inthraerythrocytic developmental cycle (IDC): 5 hours post-invasion (hpi) rings, 14 hpi mid-trophozoites, 20 hpi late trophozoites, 24 hpi mid-schizonts and 27 hpi late schizonts. The parasite culture was tightly synchronized within a 1-hour window, and harvested at the above mentioned time points. mRNA library preparation was carried out with the NEBNext Ultra II Directional RNA Library Prep Kit for Illumina, and human globin mRNA was blocked using the QIAseq FastSelect - Globin Kit. In addition, P. knowlesi line A1-H.1 was whole genome sequenced as part of the same experiment in which RNA samples for transcriptional analysis were collected. Library preparation was carried out by Novogene (Munich, Germany) using their PCR-based 'Microbial WGS' workflow (including 10 cycles of PCR amplification). 本研究提供了约氏疟原虫(Plasmodium knowlesi,P. knowlesi)A1-H.1品系在红细胞内发育周期(intraerythrocytic developmental cycle, IDC)五个时间点的批量mRNA测序数据:入侵后5小时(hpi)的环状体、14 hpi的中期滋养体、20 hpi的晚期滋养体、24 hpi的中期裂殖体以及27 hpi的晚期裂殖体。该寄生虫培养物在1小时的时间窗口内实现了严格同步化,并于上述预设时间点收获样本。mRNA文库制备采用适配Illumina测序平台的NEBNext Ultra II定向RNA文库制备试剂盒完成,同时使用QIAseq FastSelect - Globin试剂盒阻断去除人类珠蛋白mRNA。此外,本研究还对约氏疟原虫A1-H.1品系开展了全基因组测序,该测序工作作为收集转录组分析用RNA样本的同一实验的组成部分完成。全基因组文库制备由位于德国慕尼黑的诺禾致源(Novogene)完成,采用其基于PCR扩增的「微生物全基因组测序(WGS)」工作流程(包含10轮PCR扩增循环)。相关预印本可在bioRxiv平台获取:https://www.biorxiv.org/content/10.64898/2026.02.09.704036v1
