The transcriptome of leaves from two Anthurium cultivars, Oregon (Elegang, E, cold tolerant) and Fantasy love (Menghuan, MH, cold sensitive) under cold stress were sequenced by Illumina Novaseq⢠6000. Sequencing generated a total of 129.44 Gb of raw reads and an average of 7.19 Gb of reads for each sample. The analysis showed differences of transcriptomes between the control and the cold treatment samples. We identified numerous differentially expressed genes that exhibited distinct expression patterns. These genes have known or potential roles inAnthurium cold tolerance.Therefore, we are appealing candidates for further investigation of the gene expression and associated regulatory mechanisms related to the cold stress response of A. andraeanum. Overall design: The full-flowering plants were grown in the growth chambers for three days with a 12/12 h day/night cycle at 25?. For cold treatments, the plants were grown at 15? for three days, then the temperature was decreased to 6? for three days and then 4? for three days. The control plants were grown at 25? throughout the experiment. Leaves were collected at 25?(control/CK), three days after the treatments at 6? and 4? for RNAseq. Three independent experimental replicates were performed, each consisting of three independent biological replicates.

将盛花期植株置于光照/黑暗周期为12/12 h、温度25℃的人工气候箱中预培养3天。冷胁迫处理组的培养流程为:先在15℃下培养3天,随后降温至6℃培养3天,最后降至4℃继续培养3天。对照组植株在整个实验期间均维持25℃培养。分别在25℃(对照组/CK)、6℃处理3天以及4℃处理3天后采集叶片,用于RNA测序(RNAseq)。本实验共设置3次独立实验重复,每次实验重复包含3份独立生物学重复样本。
