Background and aims: The aim of this study was to analyze the molecular characteristics of adenocarcinoma of the gastroesophageal junction (AGEJ) compared to esophageal (EAC) and gastric adenocarcinomas (GCFB) from The Cancer Genome Atlas (TCGA) and Seoul National University (SNU) cohorts using next-generation sequencing. Methods: Based on mRNA expression of EAC (n=78) and GCFB (n=102) from the TCGA cohort, a molecular classification model using the Bayesian compound covariate predictor classified AGEJ/cardia (n=48) from the TCGA cohort and AGEJ/upper third gastric adenocarcinoma (n=46) from the SNU cohort into the EAC-like or GCFB-like groups. The molecular characteristics between the EAC-like and GCFB-like groups were compared. Overall design: 46 paired tumor samples-corresponding normal mucosa of SNU cohort (92 samples) were analyzed with RNA sequencing data.
背景与目的:本研究旨在通过下一代测序(next-generation sequencing)技术,对比分析癌症基因组图谱(The Cancer Genome Atlas, TCGA)队列与首尔国立大学(Seoul National University, SNU)队列中,胃食管交界处腺癌(adenocarcinoma of the gastroesophageal junction, AGEJ)与食管腺癌(esophageal adenocarcinoma, EAC)、胃腺癌(gastric adenocarcinomas, GCFB)的分子特征。
方法:基于TCGA队列中78例食管腺癌(EAC)、102例胃腺癌(GCFB)的mRNA表达数据,本研究采用贝叶斯复合协变量预测器构建的分子分类模型,将TCGA队列中48例胃食管交界处/贲门腺癌(AGEJ/cardia)样本,以及SNU队列中46例胃食管交界处腺癌/胃上三分之一腺癌样本,划分为EAC样亚型或GCFB样亚型,并对比两类亚型间的分子特征差异。
整体研究设计:对SNU队列中的46例配对肿瘤样本及其对应正常黏膜(共92例样本)开展RNA测序数据分析。
