The etiology of fibrolamellar hepatocellular carcinoma (FL-HCC), a liver cancer occurring increasingly in children to young adults, is poorly understood. We performed high-throughput sequencing on mRNA isolated from a newly developed model of FL-HCC and 4 different maturational lineages of the hepatobiliary system. Our results indicate that the transcriptome of FL-HCC is most similar to that of biliary tree stem cells. Overall design: mRNA profiles of a newly developed model of FL-HCC and 4 maturational stages of the human hepatobiliary system (biliary tree stem cells, hepatic stem cells, hepatoblasts, and adult hepatocytes) were generated by high throughput sequencing using the Illumina HiSeq 2500.
纤维板层型肝细胞癌(fibrolamellar hepatocellular carcinoma, FL-HCC)是一种在儿童至年轻成人中发病率逐渐升高的肝癌,其病因学目前仍知之甚少。本研究对一种新构建的FL-HCC模型以及肝胆系统的4种不同成熟谱系细胞所提取的mRNA进行了高通量测序(high-throughput sequencing)。研究结果显示,FL-HCC的转录组(transcriptome)与胆道树干细胞(biliary tree stem cells)的转录组最为相似。总体实验设计:本研究采用Illumina HiSeq 2500平台开展高通量测序,获得了新构建的FL-HCC模型以及人类肝胆系统4个成熟阶段(胆道树干细胞、肝干细胞(hepatic stem cells)、肝母细胞(hepatoblasts)与成人肝细胞(adult hepatocytes))的mRNA表达谱。
