Pregnancy loss is a significant problem when embryos produced in vitro are transferred to a synchronized uterus. Currently, mechanisms that underly losses of in vitro produced embryos during implantation are largely unknown. We investigated this problem using cattle (Bos taurus) as a biomedical model of conceptus attachment by analyzing transcriptome data of paired extraembryonic membrane and endometrial samples collected on gestation days 18 and 25, which spans the attachment window in cattle. Overall design: Pregnancies were initiated by either artificial insemination or the transfer of an embryo proced in vitro, then interrupted on either gestation day 18 or 25 for the collection of samples from the endometrium and conceptus.

当体外(in vitro)制备的胚胎移植至同步化子宫时,妊娠丢失是一个较为突出的问题。目前,体外制备胚胎在着床过程中发生丢失的潜在分子机制仍未被充分阐明。本研究以牛(Bos taurus)作为孕体附植的生物医学模型,通过分析妊娠第18天与第25天(覆盖牛胚胎附植窗口期)采集的成对胚外膜与子宫内膜样本的转录组数据,对该问题展开系统性探究。 实验设计:通过人工授精或体外胚胎移植建立妊娠模型,分别于妊娠第18天或第25天终止妊娠,采集子宫内膜与孕体样本。
