Purpose: Elucidate the hepatocyte-specific effects of innate antiviral response to a hepatotropic virus. Methods: Hepatocytes were isolated using a standard 2 step perfusion protocol and viable CD45 negative single cells were sorted on a Sony SH800 instrument und subsequently processed using a 10x Genomics Controller according to standard protocols. Results: Hepatocytes upregulate inflammatory genes and downregulate metabolic genes. Conclusion: Hepatocytes are important innate immune signaling platforms in the liver during viral infection. Overall design: sorted hepatocytes were processed with the 10X Genomics Chromium Single Cell and sequenced on the Illumina HiSeq 4000
研究目的:阐明针对嗜肝病毒(hepatotropic virus)的固有抗病毒应答(innate antiviral response)在肝细胞中的特异性效应。
实验方法:采用标准两步灌流方案分离肝细胞,通过Sony SH800仪器分选存活的CD45阴性单细胞,随后依照标准流程使用10x Genomics控制器完成样本处理。
实验结果:肝细胞可上调炎症基因的表达,并下调代谢基因的表达。
研究结论:病毒感染期间,肝细胞是肝脏内重要的固有免疫信号转导平台。
整体实验设计:分选所得肝细胞采用10x Genomics Chromium单细胞系统进行处理,并在Illumina HiSeq 4000测序平台上完成测序。
