mRNA m5C modification of cisplatin-resistant A549 cells

In this study, we compared the parental A549 cells with the constructed cisplatin-resistant A549 (A549/CDDP) cells at the levels of transcription and mRNA m5C methylation through RNA-Seq and RNA-BisSeq, respectively. Our data suggested that global changes occurred in cisplatin-resistant NSCLC cells and the median level of mRNA m5C modification in the A549/CDDP cells decreased compared to the A549 cells.

本研究通过RNA测序（RNA-Seq）与RNA亚硫酸氢盐测序（RNA-BisSeq），分别在转录水平及mRNA m5C甲基化水平维度，对比了亲本A549细胞与构建获得的顺铂耐药A549（A549/CDDP）细胞。研究数据表明，顺铂耐药非小细胞肺癌（Non-Small Cell Lung Cancer, NSCLC）细胞存在全局表达谱改变；且相较于亲本A549细胞，A549/CDDP细胞中mRNA m5C修饰的中位水平有所降低。
