The paired kidneys are the central organs of homeostasis for our body systems, the sophisticated structure and complex cells composition underpins kidneys' functions. Here we reported the generation of human kidney progenitor assembloid (hKPA) with expandable nephron progenitor cells (NPC) and ureteric bud (UB). These generated hKPA (in vitro) showed patterned structure and included nephron, collecting duct and interstitium. 成对肾脏是维持机体系统稳态的核心器官,其精密的结构与复杂的细胞组成是肾脏功能的核心支撑。本研究报道了由可扩增肾单位祖细胞(nephron progenitor cells, NPC)与输尿管芽(ureteric bud, UB)构建的人肾祖细胞组装体(human kidney progenitor assembloid, hKPA)。该体外构建的hKPA呈现模式化结构,包含肾单位、集合管与间质组织。实验整体设计:对培养至第25天的人肾祖细胞组装体(hKPAs)采用10x Genomics多组学(scRNA/scATAC-seq)技术开展细胞群体谱分析。
