To obtain a better perception of the molecular machinery functioning at reproductive stage which is more prone to drought stress we performed transcriptome analysis in tolerant and sensitive rice genotypes Towards this end we performed comparative transcriptome analysis of N22 and IR64 rice cultivars by transcriptome sequencing under control and drought stress conditions at the reproductive stage in both leaf and root tissues Total RNA was isolated from the leaf and root tissues of each cultivar by using TRI Reagent Two biological replicates from each cultivar were used for transcriptome sequencing The cDNA libraries were prepared individually for each sample following a series of procedures using mRNASeq Sample Prep Kit from Illumina following the manufacturers instructions for paired-end sequencing A total of eight transcriptome libraries were prepared from leaf and root tissues from the two rice cultivars grown under control and drought stress, and sequenced using Illumina platform
为更深入解析易受干旱胁迫影响的水稻生殖阶段分子调控机制,我们对耐旱与敏感水稻基因型开展了转录组(transcriptome)分析。为此,我们针对N22与IR64两个水稻品种,在生殖阶段的叶片和根系组织中设置对照与干旱胁迫处理,通过转录组测序开展比较转录组分析。采用TRI Reagent试剂提取各品种叶片与根系组织的总RNA,每个品种设置两个生物学重复用于转录组测序。按照Illumina公司的mRNASeq Sample Prep Kit试剂盒的厂商操作说明,针对双端测序需求,通过一系列实验步骤为每个样本单独构建cDNA文库。本研究共为两个水稻品种在对照与干旱胁迫条件下的叶片和根系组织构建了8个转录组文库,并通过Illumina测序平台完成测序。
