We analyed the gene expression profiles after knocking down MYCN or TFAP4. Results showed that transcription factor MYCN and TFAP4 commonly regulats a subset of genes that may contribute to neuroblastoma cells proliferation and migration. Overall design: Neuroblastoma BE(2)-C cells were transfected with control siRNA, MYCN siRNA1, MYCN siRNA2, TFAP4 siRNA1 and TFAP4 siRNA2, respectively. Total RNA was extracted from cells 30 hours after each transfection and subjected to gene expression study by Affymetrix microarray.
本研究分析了MYCN或TFAP4基因敲低后的基因表达谱。结果表明,转录因子(transcription factor)MYCN与TFAP4可共同调控一组可能参与神经母细胞瘤细胞增殖与迁移的基因。实验设计概述:将神经母细胞瘤BE(2)-C细胞分别转染阴性对照小干扰RNA(siRNA)、MYCN siRNA1、MYCN siRNA2、TFAP4 siRNA1及TFAP4 siRNA2,于每轮转染后30小时提取细胞总RNA,通过Affymetrix基因芯片开展基因表达分析。
