We observed the expression profile of the total mRNA of wild-type Thermus thermophilus HB8 strain grown in a rich (TR) medium at 70°C. Keywords: time course Three wild-type T. thermophilus HB8 strains were pre-cultured at 70°C for 16 h in 3 ml of TR medium containing 0.8% polypeptone, 0.4% yeast extract, 0.2% NaCl, 0.4 mM CaCl2, and 0.4 mM MgCl2, which was adjusted to pH 7.2 with NaOH. The cells (2 ml) were inoculated into 1 liter of the same medium and then cultivated at 70°C. Cells were collected after 180, 240, 300, 360, 420, 480, 540, 600, 680, and 760 min, and then crude RNA was extracted. The expression of each mRNA at each time point was analyzed on a GeneChip as described under Sample Description Sheet of each sample in this website.

本研究对在70℃条件下于丰富培养基(TR培养基)中培养的野生型嗜热栖热菌(Thermus thermophilus)HB8菌株的总mRNA表达谱进行了观测。关键词:时间进程。取3株野生型嗜热栖热菌HB8,于3 mL TR培养基中70℃预培养16 h。该TR培养基含0.8%胰蛋白胨、0.4%酵母提取物、0.2%氯化钠、0.4 mM氯化钙及0.4 mM氯化镁,经氢氧化钠溶液调节pH至7.2。取2 mL上述菌液接种至1 L相同培养基中,随后于70℃条件下继续培养。分别于培养至180、240、300、360、420、480、540、600、680及760 min时收集菌体,随后提取粗总RNA。针对各时间点下的各mRNA表达水平,采用GeneChip(基因芯片)进行分析,具体操作参照本网站各样本的样本描述文档。
