Mouse ES cells were stably transduced with a lentivirus expressing either wild-type KBP or the stable mutant KBP(KK/RR) and maintained in self-renewing growth conditions. RNA-seq was performed to assess mRNA expression differences caused by the stabilization of KBP. 6 samples [a triplicate set for ES cells expressing wild-type KBP and a triplicate set expressing KBP(KK/RR)] were analyzed.
将小鼠胚胎干细胞(Mouse ES Cells)分别采用表达野生型KBP与稳定突变体KBP(KK/RR)的慢病毒进行稳定转导,并维持于自我更新培养条件下。通过RNA测序(RNA-seq)技术,评估KBP稳定化所引发的mRNA表达差异。本实验共分析6份样本,其中表达野生型KBP的胚胎干细胞与表达KBP(KK/RR)的胚胎干细胞各设置3个生物学重复样本组。
